FBS Thawing & Aliquoting Protocol

Step-by-step guide for thawing, aliquoting, and storing Fetal Bovine Serum for optimal cell culture performance.

Steps 1–2

Thaw at 37°C — Never Higher

Water bath method (preferred): Place frozen bottle in a 37°C water bath for 2–5 minutes with gentle swirling. Remove as soon as a small ice crystal remains.

Refrigerator method: Transfer from −20°C/−80°C to 2–8°C refrigerator overnight. Best for minimizing thermal shock to proteins.

Precipitate is normal. Flocculent material after thawing is fibrin and lipoproteins — it does not indicate contamination and won't affect cell culture performance.

Never thaw FBS in a microwave or above 37°C. Excessive heat denatures growth factors.

Step 3

Divide Into Working Volumes

Work in a biological safety cabinet (BSC). Spray bottle with 70% ethanol before opening.

Recommended aliquot volumes:

  • 15 mL — for 150 mL media at 10% FBS
  • 25 mL — for 250 mL media at 10% FBS
  • 50 mL — for 500 mL media at 10% FBS

Label each tube with lot number, date, and volume. Critical for lot traceability and troubleshooting.

Steps 4–5

Freeze Aliquots, Thaw Once

Storage:

  • −20°C — routine storage, up to 12 months
  • −80°C — long-term storage, 2+ years
  • Avoid frost-free freezers — auto-defrost cycles cause temperature fluctuations

Using aliquots: Thaw one aliquot at 37°C per Step 1. Add to media. Supplemented media stores 2–4 weeks at 2–8°C.

Never refreeze a thawed aliquot. Each freeze-thaw cycle degrades growth factors by 5–10%.

Not recommended. Room temperature thawing is slow and uneven, increasing the risk of protein degradation and bacterial growth. Always use a 37°C water bath (2–5 minutes) or a 2–8°C refrigerator (overnight).
